Development and evaluation of an enzyme-linked immunosorbent assay using recombinant p23 for the detection of antibodies to Cryptosporidium parvum in cattle OAK

An enzyme-linked immunosorbent assay (ELISA) based on the recombinant p23 of Cryptosporidium parvum was established for the detection of antibodies against C. parvum in cattle. The sensitivity and specificity of the ELISA were evaluated with the standard indirect fluorescent antibody test (IFAT) using sporozoites as antigens. Of 77 bovine sera collected from China, 20 (26.0%) were identified as positive by the IFAT. The same samples were tested with the ELISA. The optical densities at 415 nm were compared to the IFAT results and statistically analyzed to designate a provisional cut-off point. As a result, the cut-off point was concluded to be 0.08, which was considered to be the best condition in the light of its sensitivity (80%) and specificity (73.7%)....

Diagnosis of Babesia caballi Infection in Horses by Polymerase Chain Reaction OAK

A set of primers were designed according to the published sequence of the gene encoding a rhoptry protein of Babesia caballi, and used to amplify parasite DNA from the blood samples obtained from carrier horses by polymerase chain reaction(PCR)method.The PCR method was sensitive enough to detect parasite DNA from 2.5 μl blood sample with a parasitemia of 0.000001%. The PCR method was compared with fluorescent antibody test(IFAT) in order to evaluate the diagnosis effciency for B. caball infection in horses. Of 142 field samples from Mongolia, 28(20%) and 96(69%)samples were identified positively by PCR and IFAT, respectively. Genetic diversity and antigenicity variation of Babesia bovis merozoite surface antigen-1 (MSA-1) in Thailand OAK

Babesia bovis, an intraerythrocytic protozoan parasite, causes severe clinical disease in cattle worldwide. The genetic diversity of parasite antigens often results in different immune profiles in infected animals, hindering efforts to develop immune control methodologies against the B. bovis infection. In this study, we analyzed the genetic diversity of the merozoite surface antigen-1 (msa-1) gene using 162 B. bovis-positive blood DNA samples sourced from cattle populations reared in different geographical regions of Thailand. Recently,we reported the in vitro growth inhibitory effects of the imidazole derivatives,clotrimazole(CLT) and ketoconazole (KC),and the herbicide,clodinafop-propargyl(CP),against bovine and equine Babesia parasites. Additionally,in the bovine species,several combined applications of the CLT,KC,and CP were reported to significantly increase the inhibitory effects. In this study,similar combination tests were carried out in order to confirm possible synergistic effects in the equine species,Babesia caballi and Babesia equi. Combinations of CLT/KC, CLT/CP,and CLT/KC/CP exerted a significantly enhanced growth inhibitory efficacy in B. caballi,in contrast to B. equi,where no synergistic effects were observed in any of the combinations. In vitro inhibitory effect of fosmidomycin on the asexual growth of Babesia bovis and Babesia bigemina OAK

The in vitro inhibitory effect of fosmidomycin was evaluated against the asexual growth of bovine Babesia parasites (Babesia bovis and Babesia bigemina). In the cultures with these parasites, severe reduction of parasite growth was observed by the addition of 1.25 μg/ml of the final drug concentration. Under these conditions, B. bigemina was completely cleared until the third day of cultivation. On the other hand, B. bovis was completely eliminated on the fourth day when the concentration was 6.25 μg/ml in the culture. These parasites failed to grow again when a drug-free medium was substituted for the subsequent cultivation. Inhibitory Effects of Pepstatin A and Mefloquine on the Growth of Babesia Parasites OAK

We evaluated the inhibitory effects of pepstatin A and mefloquine on the in vitro and in vivo growths of Babesia parasites. The in vitro growth of Babesia bovis, B. bigemina, B. caballi, and B. equi was significantly inhibited (P < 0.05) by micromolar concentrations of pepstatin A (50% inhibitory concentrations = 38.5, 36.5, 17.6, and 18.1 μM, respectively) and mefloquine (50% inhibitory concentrations = 59.7, 56.7, 20.7, and 4 μM, respectively). Furthermore, both reagents either alone at a concentration of 5 mg/kg or in combinations (2.5/2.5 and 5/5 mg/kg) for 10 days significantly inhibited the in vivo growth of B. microti in mice. Molecular and Immunological Characterization of a Novel 32-kDa Secreted Protein of Babesia microti OAK

A cDNA encoding the Babesia microti 32-kDa protein was identified by serological immunoscreening of a cDNA expression library and designated as BmP32. The full length of BmP32 contains an open reading frame of 918 base pairs consisting of 306 amino acids having a significant homology with B. microti secreted antigen 1. Antiserum raised against recombinant protein (rBmP32) specifically reacted with a 32-kDa native protein of the parasite lysate using western blot analysis. Background: Babesia canis is an apicomplexan tick-transmitted hemoprotozoan responsible for causing canine babesiosis in Europe and west Asia. Despite its importance, there is no known rapid diagnostic kit detection of B. canis infection in dogs. Here we report on an epidemiological study regarding the molecular prevalence of different genotypes of Theileria orientalis present among domestic cattle and water buffalo populations bred in Thailand. A phylogenetic analysis based on the parasitic gene encoding a major piroplasm surface protein revealed the presence of 5 genotypes (Types 1, 3, 5, 7, and N-3) in cattle and 7 genotypes (Types 1, 3, 4, 5, 7, N-2, and N-3) in water buffaloes. Preliminary evaluation of oligomannose-coated liposome vaccines against lethal protozoan infections in mice OAK

The oligomannose-coated liposome (OML) vaccine is known to induce cellular immunity specific for the encapsulated antigen in immunized mice. In the present study, we preliminarily evaluated the effect of the OML vaccine encapsulating the soluble protozoan lysate of Toxoplasma gondii, Trypanosoma brucei gambiense, or Babesia rodhaini on the corresponding protozoan infections in mice. After the challenge of T. gondii, the OML vaccine group avoided the high mortality resulting from acute infection that was dominantly observed in other control groups. A heterologous prime-boost immunization regime with priming DNA followed by recombinant vaccinia virus expressing relevant antigens has been shown to induce effective immune responses against several infectious pathogens. In this study, we constructed a recombinant plasmid and vaccinia virus, both of which expressed P50 of Babesia gibsoni, to investigate the immunogenicity and protective efficacy of a heterologous prime-boost immunization against canine babesiosis.
